    ABSTRACT

    Studies were performed on the production of extracellular polysaccharide in submerged culture of Agaricus brasiliensis Wasser et al. and the effects of different carbon and nitrogen sources individually and in combinations on production. The polysaccharide produced was evaluated for its antitumor action against Sarcoma 180. Glucose and yeast extract gave the best combination of carbon and nitrogen sources for the polysaccharide production. The biomass yield and polysaccharide produced were 222.3 and 10.867 mg/50 mL (dry wt), respectively, at the concentration of 20 g/L glucose and 2 g/L yeast extract with the medium containing MgSO4Ч7H2O (0.1%) and K2HPO4Ч3H2O (0.1%) at pH 6.0 and 32°C after the 9 days' static cultivation. The extracellular polysaccharide showed strong antitumor activity against Sarcoma 180. Complete regression ratio of 50% and suppression ratio percentage of 72.19% were observed.
