    ABSTRACT

    Agaricus brasiliensis is included in many kinds of health foods. Both the fruit body and mycelium, cultured in various countries from various strains and using various production processes, are supplied. In the present study, we planned to compare the chemical properties and biological activities of A. brasiliensis cultured under different conditions, i.e., indoors in a house (IC) and outside on a field (OC), using the same strain and the same production process. Polysaccharide fractions prepared from IC and OC had similar structural features and biological activity, as assessed by component sugar analysis, nuclear magnetic resonance, and antitumor activity in vivo. In contrast, the protein fractions were qualitatively different, i.e., specific protein spots were detected by two-dimensional polyacrylamide gel electrophoresis, and the enzyme activities, especially laccase, polyphenoloxidase, peroxidase, and beta-glucanase activities, were different. These facts strongly suggested that the culture conditions of medicinal mushrooms are keys for their pharmacological activity assessment.
